The Cocktail Report (sound really smart around your friends):

  • AMPK is an enzyme that senses when a cell is low on fuel and tells it to burn reserves instead of building and storing.

  • Metformin nudges AMPK indirectly, so nobody could say for sure whether AMPK itself drives its anti-aging effects in animals.

  • A direct activator called 991 lengthened life in fruit flies, roundworms and fission yeast, published in Aging Cell on September 29, 2026.

  • In worms and yeast lacking AMPK, the drug gave no lifespan gain, which points straight at the enzyme.

  • Treated flies carried less body fat while eating just as much as untreated flies.

Every time you exercise or skip a meal, an enzyme called AMPK notices the dip in fuel and changes what your cells do. A team led by the MRC Laboratory of Medical Sciences in London has now shown that switching AMPK on with a drug, and nothing else, makes three kinds of organisms live longer.

The Medical Research Council's announcement said the gains reached more than 25% in some cases. The paper itself, a short report in Aging Cell, was first published September 29, 2026.

Why a "direct" activator matters
AMPK (adenosine monophosphate-activated protein kinase) works like a phone's power-saving mode. When energy runs low, it shuts down costly jobs such as building proteins and storing fat, and it turns up fat and sugar burning.

If you take metformin, or have seen it sold as an anti-aging pill, you have a stake in this question. Metformin raises AMPK activity only by first disturbing the cell's energy balance, and it acts on mitochondria, lysosomes and even gut bacteria along the way.

That mix made it hard to say which effect mattered. The compound 991 instead locks into a pocket on AMPK called the ADaM site and turns the enzyme on directly.

What the drug did
In fruit flies, about 150 females per group, 991 in the diet extended life at both 10 and 100 micromolar doses. Treated flies stored less triglyceride, the main form of body fat, yet ate the same amount and laid the same number of eggs.

In roundworms, about 110 per group, 991 extended life whether the worms were fed one strain of E. coli or another. Fission yeast, a single-celled fungus, also survived longer at 10 micromolar.

The test that points at AMPK
The team then gave 991 to worms and yeast whose AMPK gene had been knocked out. The worms got no lifespan gain, and the yeast actually died sooner.

That result matters because it rules out a side target. The longer life ran through AMPK.

A first look in mice
The team also gave male mice 991 packed in tiny biodegradable particles, injected three times a week for 3 weeks. Liver proteins in the treated mice, 3 per group, shifted toward more energy production, less mTOR (a growth signal tied to aging), and more mitochondria, the cell's powerhouses.

No mouse lifespan was measured. The authors say mouse lifespan studies are now warranted.

What this means for you
Drug companies already make stronger cousins of 991 that bind the same pocket. One of them, PXL-770, has been tested in human trials with good safety, and patients with fatty liver disease had better blood fats after 4 weeks.

Exercise and fasting already switch AMPK on in your own body. That remains the version you can use today.

What these results do and do not show
To be frank, worms live about three weeks and flies about three months, compared with roughly three years for mice. None of that tells us what happens in a mammal, and this paper has no mammal lifespan data.

Dose cut both ways: 250 micromolar shortened fly life, and 100 micromolar shortened yeast life.

Co-author Charalampos Rallis of Queen Mary University of London put it plainly: "I would not want anyone to reach for a supplement on the back of this. Too much of the drug shortened life in our experiments.

Why Should You Care?
This is the clearest evidence so far that AMPK itself, not some side effect of metformin, can slow aging in animals. The next step is mice, and the tools to run that test already exist.
